Influence of sediment composition on PAH toxicity using zebrafish (Danio rerio) and Japanese medaka (Oryzias latipes) embryo-larval assays ArchiMer
Perrichon, Prescilla; Le Bihanic, Florane; Bustamante, Paco; Le Menach, Karyn; Budzinski, Helene; Cachot, Jerome; Cousin, Xavier.
Due to hydrophobic and persistent properties, polycyclic aromatic hydrocarbons (PAHs) have a high capacity to accumulate in sediment. Sediment quality criteria, for the assessment of habitat quality and risk for aquatic life, include understanding the fate and effects of PAHs. In the context of European regulation (REACH and Water Framework Directive), the first objective was to assess the influence of sediment composition on the toxicity of two model PAHs, benzo[a]pyrene and fluoranthene using 10-day zebrafish embryo-larval assay. This procedure was undertaken with an artificial sediment in order to limit natural sediment variability. A suitable sediment composition might be then validated for zebrafish and proposed in a new OECD guideline for chemicals...

Long-term disruption of growth, reproduction, and behavior after embryonic exposure of zebrafish to PAH-spiked sediment ArchiMer
Vignet, Caroline; Devier, Marie-hélène; Le Menach, Karyn; Lyphout, Laura; Potier, Jeremy; Cachot, Jerome; Budzinski, Helene; Begout, Marie-laure; Cousin, Xavier.
A natural sediment spiked with three individual polycyclic aromatic hydrocarbons (PAHs; pyrene, phenanthrene and benzo[a]pyrene) was used to expose zebrafish embryos and larvae during 4 days. The total PAH concentration was 4.4 μg g−1 which is in the range found in sediment from contaminated areas. Quantification of metabolites in the larvae after exposure confirmed the actual contamination of the larvae and indicated an active metabolism especially for pyrene and benzo[a]pyrene. After a transfer in a clean medium, the larvae were reared to adulthood and evaluated for survival, growth, reproduction, and behavior. Measured endpoints revealed a late disruption of growth (appearing at 5 months) and a trend toward a lower reproductive ability. Adults of...

Parental trophic exposure to three aromatic fractions of polycyclic aromatic hydrocarbons in the zebrafish: Consequences for the offspring ArchiMer
Perrichon, Prescilla; Akcha, Farida; Le Menach, Karyn; Goubeau, Manon; Budzinski, Helene; Cousin, Xavier; Bustamante, Paco.
In recent decades, PAH emissions due to extensive anthropogenic activities have risen sharply causing considerable pollution of aquatic ecosystems. This pollution represents a threat for organisms, among them are fish. Consequently, prenatal stress can have important repercussions, and may impact survival and population recruitment. To investigate this point, eggs were collected from zebrafish exposed during 6 months by trophic route to three aromatic fractions from two different origins, pyrolytic (PY) and petrogenic (light (BAL) and heavy (HFO) fractions) sources. Chronic dietary exposure of the parents was performed at environmentally relevant concentrations (0.3 ×, 1 × and 3 ×; 1 × represents an environmental concentration measured in French estuary)....

Pharmaceuticals, alkylphenols and pesticides in Mediterranean coastal waters: Results from a pilot survey using passive samplers ArchiMer
Munaron, Dominique; Tapie, Nathalie; Budzinski, Helene; Andral, Bruno; Gonzalez, Jean-louis.
21 pharmaceuticals, 6 alkylphenols and 27 hydrophilic pesticides and biocides were investigated using polar organic contaminant integrative samplers (POCIS) during a large-scale study of contamination of French Mediterranean coastal waters. Marine and transitional water-bodies, defined under the EU Water Framework Directive were monitored. Our results show that the French Mediterranean coastal waters were contaminated with a large range of emerging contaminants, detected at low concentrations during the summer season. Caffeine, carbamazepine, theophilline and terbutaline were detected with a detection frequency higher than 83% in the coastal waters sampled, 4-nonylphenol (4-NP), 4-tert-octylphenol (4-OP) and 4-nonylphenol diethoxylate (NP2EO) were detected...

Predicted environmental concentrations of carbamazepine, oxcarbazepine and their main metabolites in a coastal system ArchiMer
Fenet, Helene; Arpin-pont, Lauren; Vanhoutte Brunier, Alice; Munaron, Dominique; Fiandrino, Annie; Hilaire-buys, Dominique; Mathieu, Olivier; Budzinski, Helene; Chiron, Serge; Boillot, Clotilde; Gomez, Elena.
Pharmaceuticals are widely released in aquatic environment through treated wastewaters. They reach coastal zone indirectly via streams or directly though marine outfalls however data concerning this contamination in coastal waters are scarce. Environmental Risk Assessment (ERA) of pharmaceuticals have been conducted mostly in surface waters and has not been performed in coastal zone. The first step of ERA is to evaluate the exposure through predictive environmental concentration (PEC) values. The aim of this study was to predict the occurrence of some pharmaceuticals in a coastal aera subjected to treated wastewater (TWW) reject through a marine outfall (Fig 2.). Among pharmaceuticals, Carbamazepine, Oxcarbazepine and their main metabolites (Fig 1.) were...

PROJET PEPS Guyane - Evaluation de la contamination chimique des eaux guyanaises par les techniques d'échantillonnage passif. Application et soutien à la mise en place de la DCE. Campagne Octobre 2009 ArchiMer
Gonzalez, Jean-louis; Tapie, Nathalie; Guyomarch, Julien; Budzinski, Helene; Van Ganse, Sophie; Luglia, Mathieu.

ArchiMer
Gonzalez, Jean-louis; Tapie, Nathalie; Guyomarch, Julien; Budzinski, Helene.

Gonzalez, Jean-louis; Turquet, Jean; Cambert, Harold; Budzinski, Helene; Tapie, Nathalie; Guyomarch, Julien; Andral, Bruno.

While they had previously presented a hight stability, unlike in many coastal sites worldwide, eelgrass beds (Zostera noltii and Zostera marina) of Arcachon Bay have declined sharply since the 2000s. Various causes have been invoked to explain this phenomenon: climate anomalies, disease, over-grazing by herbivorous birds, contamination by herbicides, mechanical destruction during clam fishing. A multidisciplinary study was set up to study the possible effect of these factors, by putting into perspective the history of the seagrass beds and the temporal evolution of these parameters. After a chapter dealing with recent trends (2007-2010) of seagrasses in the Bay of Arcachon, the results relative to the various hypotheses are presented. These results allow...

Simulated conservative tracer as a proxy for S-metolachlor concentration predictions compared to POCIS measurements in Arcachon Bay ArchiMer
Fauvelle, Vincent; Belles, Angel; Budzinski, Helene; Mazzella, Nicolas; Plus, Martin.
The work presented here aims at comparing monitoring of S-metolachlor, the major pesticide in use in the Arcachon Bay (South West of France, transitional coastal area), by chemical analysis (monthly passive sampling) and contaminant dissipation modeling from sources (Mars-2D model). The global strategy consisted in i) identifying the major sources of S-metolachlor to the Bay, ii) monitoring these sources for 12 months, and iii) comparing modeled data in the Bay based on measured inputs, to chemical measurements made inside the Bay along with the 12-month source monitoring. Results first showed that the major S-metolachlor surface inputs to the Arcachon Bay are mainly from one single source. Modeled and measured data were in good agreement at 5 sites in the...

Pesticides represent a major proportion of the chemical pollutants detected in French coastal waters and hence a significant environmental risk with regards to marine organisms. Commercially-raised bivalves are particularly exposed to pollutants, among them pesticides, as shellfish farming zones are subject to considerable pressure from agricultural activities on the mainland. The aims of this study were to determine (1) the genotoxic effects of diuron exposure on oyster genitors and (2) the possible transmission of damaged DNA to offspring and its repercussions on oyster fitness. To investigate these points, oysters were exposed to concentrations of diuron close to those detected in the Marennes-Oleron Basin (two 7-day exposure pulses at 0.4 and 0.6 μg...

The influence of natural dissolved organic matter on herbicide toxicity to marine microalgae is species-dependent ArchiMer
Coquille, Nathalie; Menard, Dominique; Rouxel, Julien; Dupraz, Valentin; Eon, Melissa; Pardon, Patrick; Budzinski, Helene; Morin, Soizic; Parlanti, Edith; Stachowski-haberkorn, Sabine.
Microalgae, which are the foundation of aquatic food webs, may be the indirect target of herbicides used for agricultural and urban applications. Microalgae also interact with other compounds from their environment, such as natural dissolved organic matter (DOM), which can itself interact with herbicides. This study aimed to evaluate the influence of natural DOM on the toxicity of three herbicides (diuron, irgarol and S-metolachlor), singly and in ternary mixtures, to two marine microalgae, Chaetoceros calcitrans and Tetraselmis suecica, in monospecific, non-axenic cultures. Effects on growth, photosynthetic efficiency (Ф’M) and relative lipid content were evaluated. The chemical environment (herbicide and nutrient concentrations, dissolved organic carbon...

Toxicity assessment of water-accommodated fractions from two different oils using a zebrafish (Danio rerio) embryo-larval bioassay with a multilevel approach ArchiMer
Perrichon, Prescilla; Le Menach, Karyn; Akcha, Farida; Cachot, Jerome; Budzinski, Helene; Bustamante, Paco.
Petroleum compounds from chronic discharges and oil spills represent an important source of environmental pollution. To better understand the deleterious effects of these compounds, the toxicity of water-accommodated fractions (WAF) from two different oils (brut Arabian Light and Erika heavy fuel oils) were used in this study. Zebrafish embryos (Danio rerio) were exposed during 96 h at three WAF concentrations (1, 10 and 100% for Arabian Light and 10, 50 and 100% for Erika) in order to cover a wide range of polycyclic aromatic hydrocarbon (PAH) concentrations, representative of the levels found after environmental oil spills. Several endpoints were recorded at different levels of biological organization, including lethal endpoints, morphological...
